This might be cheating (because it's actually 4 fics), but my vote goes to the Mr. William series by Pepperlandgirl.

So not fair because it changes! Right now, today, I'd go with "Just Another Saturday Night" by Poshcat over at AllAboutSpike. There's a rollicking dynamic to it I find enchanting and I feel it informs the S6 relationship. Right now. Today.

I have lots, but the one I can go back to and re-read and re-read is 'The Ultimate Betrayal' by jamies_lady. It hits all the spots for me! We have angst, horror, fighting, saving the world, love, romance, intimacy and humor with great character development. All characters present but it is a step away from the 'norm' with a great plot but enough of the 'norm' to know it is spuffy as it should be.

The Bittersweets Series by Herself. Hands down my favorite spuffy fic of all time. Bonus points for being HUGE and for all of its parts being equally magnificent. Well written, well plotted, interesting and like nothing else out there -- nothing compares.

The Origins Series by Niamh. Has to be the BEST Spike characterization ever...and yeah, it's got it all. Great ensemble, romance, and action. It's truly incredible.

Summary: Angel and Spike are members of a gangster type family ruled over by Masters (in NY) Angel runs the LA branch of the 'family' and after an affair with Buffy he disappears. Masters is furious and is determined to bring his wayward child back to be punished. Spike and Dru are lovers and have been living in NY working for Masters but when Angel vanished he took Drusilla with him...Spike is enraged and seeking revenge...then he discovers Angel's one true love (Buffy) and the perfect revenge is within his grasp...this is a Spike and Buffy story...how hate and revenge turn into lust and desire and finally into trust and love...
